Summary, etc. Xylooligosaccharides are non-digestible food ingredients that preferentially stimulate the growth of probiotics. Xylooligosaccharides can be used as prebiotics as well as functional food. Xylooligosaccharides have many beneficial effects and generally they are produced from xylan by enzymatic hydrolysis. Xylan is one of the main structural components in plant cell walls. Also it is the most abundant hemicellulose in nature. Generally, xylan can be considered as an agricultural waste. However, due to upcoming environmental changes, and the excess of residues generated by industrial process, sustainable methods are being applied in the waste management. The hemicellulose in sugarcane bagasse is rich in xylan and can be used to produce xylose, xylitol, second-generation ethanol and xylooligosaccharides. Present study investigated the alkaline extraction of sugarcane bagasse (Saccharum officinarum) with application of ultrasound in production of xylan, followed by the enzymatic hydrolysis by enzyme xylanase refined from Trichoderma reesei to produce xylooligosaccharides. The optimized conditions for xylan extraction from sugarcane bagasse were 16% KOH for 60 min of extraction time. The yield at that condition was 11.47±0.99 % and 10.1±0.1% for extraction with and without ultrasound respectively. According to statistical analysis, ultrasound application, extraction time and concentration of KOH indicated a significant effect on the xylan recovery. Furthermore the extracted xylan was explored to develop the prebiotic xylooligosaccharide. In enzymatic hydrolysis process, hydrolysis for 12 hours produced xylooligosaccharides with a maximum yield of 0.22 mg/mL. For the effect of xylooligosaccharides on probiotics, the growth of lactic acid bacteria Lactobacillus plantarum was investigated. Lactobacillus plantarum was tested for utilization of xylooligosaccharides derived from sugarcane bagasse and the growth was indicated by the increase in optical density, variation of the pH and viable cell count.
